Ontario Provincial Police will have both westbound lanes of Hwy 401 closed near Napanee, starting at 6 a.m. on Thursday.
A transport truck crashed, just east of Palace Road, at 4:11 a.m. Wednesday.
Police say complete clean-up and recovery of the truck will require both westbound lanes to be closed.
The tractor trailer lost control and damaged approximately 500 m of guard rail before tipping onto its side in the ditch.
The driver was taken to hospital and treated for minor injuries.
James Woolley, 50, of St. Thomas is being charged with careless driving.
The Hwy 401 closure is expected to take between six and eight hours.
An Emergency Detour Route (EDR) has been set up to deal with the closure:
Exit Hwy 401 at County Rd #4 (exit #593)
South on Cty Rd 4 to County Rd 2.
West on Cty 2 to Palace Road
North on Palace Road to re-enter Hwy 401 west (MM 582).
OPP add that construction underway on other roadways in the area of the EDR will create significant delays should any drivers opt to deviate from the listed detour. Police strongly encourage everyone to use the EDR as it will be the most efficient way through the area of the highway closure.
